Biological Background: N4BP1 Function and Localization
- N4BP1 (NEDD4-binding protein 1) is a potent suppressor of cytokine production and a key negative regulator of innate immune signaling, limiting inflammatory responses to minor TLR stimuli.
- Acts as a restriction factor against HIV-1 by binding viral mRNAs and degrading them through its intrinsic ribonuclease activity. Related references: PMID:31133753
- Upon infection with more threatening pathogens, N4BP1 is cleaved by CASP8 downstream of TLR3 or TLR4, leading to its inactivation and allowing full cytokine production.
- Inhibits the E3 ubiquitin-protein ligase ITCH by competing for substrate binding via the second WW domain of ITCH, impairing ubiquitination.
- Localizes to the cytoplasm, cytosol, nucleus, nucleolus, and PML nuclear bodies, indicating multifunctional cellular distribution.
- Detected in a wide range of tissues, including heart, lung, brain, liver, skeletal muscle, pancreas, kidney, spleen, testis, and ovary.
- Keywords from UniProt highlight its roles as a hydrolase, nuclease, RNA-binding protein, and phosphoprotein involved in innate immunity.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 190–590 of mouse N4BP1, a region containing the NYN (RNase) domain and part of the ITCH-interacting region. This design is expected to produce an antibody that recognizes full-length N4BP1 in denaturing and native conditions.
- For Western blot, consider a starting dilution range of 1:500–1:2000 and validate with relevant positive controls (e.g., human or mouse cell lysates known to express N4BP1).
- In immunofluorescence/ICC, confirm nuclear and cytoplasmic localization with appropriate fixation and permeabilization protocols; inclusion of PML body markers may aid co-localization studies.
- The antibody is validated for ELISA, potentially suitable for capture or detection in sandwich assays; optimize coating and detection conditions for your specific immunoassay format.
- Cross-reactivity with human and mouse N4BP1 is specified; however, verify specificity in your experimental system, especially if working with samples from other species.
