Biological Background: N-Cadherin Function and Localization
- Cadherin-2 (N-Cadherin, CDw325) is a calcium-dependent cell adhesion protein that preferentially mediates homotypic cell-cell adhesion via dimerization with CDH2 chains from adjacent cells, contributing to the sorting of heterogeneous cell types.
- It acts as a regulator of neural stem cell quiescence by mediating anchorage of neural stem cells to ependymocytes in the adult subependymal zone; cleavage by MMP24 affects this anchorage, modulating quiescence.
- CDH2 plays a role in cell-to-cell junction formation between pancreatic beta cells and neural crest stem (NCS) cells, promoting process formation by NCS cells.
- Required for proper neurite branching and pre- and postsynaptic organization; may regulate dendritic spine density in hippocampal neurons.
- May promote axon outgrowth and motor fiber repair via DSP-mediated recruitment to outgrowth tips.
- Subcellularly localized to the cell membrane, sarcolemma, cell junctions (adherens junctions, desmosomes), cell surface, and axon projections.
