Biological Background: MZB1 Function and Localization
- MZB1, also known as pERp1, MEDA-7, and PACAP, is a marginal zone B- and B1-cell-specific protein encoded by the MZB1 gene. Isoform 2 is generated by alternative splicing and may regulate apoptosis.
- Acts as a molecular chaperone or oxidoreductase, associating with IgM heavy and light chains to promote IgM assembly and secretion; it displays low oxidoreductase activity (By similarity).
- Functions as a hormone-regulated adipokine/pro-inflammatory cytokine implicated in chronic inflammation, cellular expansion, and blunted insulin response in adipocytes, potentially contributing to insulin resistance.
- Diversifies peripheral B-cell functions by regulating Ca²⁺ stores, antibody secretion, and integrin activation.
- Localizes predominantly to the endoplasmic reticulum lumen, but is also found secreted and in the cytoplasm.
- Widely expressed, with highest levels in adult brain, small intestine, thymus, and spleen; expression is frequently lower in intestinal-type gastric cancer, and in obese patients, more abundant in omental than subcutaneous fat.
- Key structural features include a signal peptide, disulfide bonds, and a determined three-dimensional structure.
Experimental Guidance and Technical Tips
- For Western blot, target detection at ~21 kDa under reducing conditions is typical. Consider using a positive control such as lysates from lymphoid tissues or MZB1-expressing cell lines.
- For immunohistochemistry (IHC-P), antigen retrieval with citrate or EDTA buffer may enhance signal due to ER localization. Validate in appropriate tissue sections (e.g., spleen, thymus).
- For ELISA, the antibody can be used as either capture or detection reagent in sandwich assays, paired with a matched monoclonal antibody if available.
- Cross-reactivity with mouse and rat is confirmed; verify performance in your specific sample system.
