Biological Background: Myocardin Function and Localization
- Myocardin (MYOCD) is a smooth muscle and cardiac muscle-specific transcriptional cofactor that acts together with serum response factor (SRF) to regulate genes via CArG box DNA sequences, playing crucial roles in cardiogenesis, urinary bladder development, and smooth muscle cell differentiation (by similarity).
- It positively regulates transcription of genes involved in vascular smooth muscle contraction (by similarity).
- Subcellular localization is predominantly nuclear, consistent with its role as a transcription factor.
- Expressed in heart, aorta, bladder, and various smooth muscle-containing tissues including stomach, small intestine, colon, lung, placenta, and uterus; very faint expression in prostate and skeletal muscle. Related references: PMID:12640126 PMID:20385216
- The protein features a coiled-coil domain and undergoes post-translational modifications including phosphorylation and ubiquitin-like conjugation. Alternative splicing may generate multiple isoforms.

- The immunogen is a synthetic peptide corresponding to amino acids 680–780 of human MYOCD. For western blotting (WB), a band around 102 kDa is expected; consider using nuclear-enriched protein extracts to improve detection sensitivity.
- This polyclonal antibody is applicable for immunohistochemistry (IHC-P) on paraffin-embedded tissues. Heart, aorta, or bladder sections from human or rat are positive controls; validate staining specificity in your own system.
- For ELISA, the peptide-based immunogen may provide good reactivity, but assay conditions should be optimized experimentally. Cross-reactivity with rat MYOCD is reported, making it suitable for rodent model studies.
