Biological Background: MYO1C Function and Localization
- MYO1C encodes unconventional myosin-Ic, also known as myosin I beta (MMI-beta, MMIb), an actin-based motor molecule with ATPase activity.
- Its tail domain mediates binding to phosphoinositides and links the actin cytoskeleton to cellular membranes, enabling intracellular movements such as glucose transporter (GLUT4) vesicle recycling in response to insulin.
- In hair cells of the inner ear, MYO1C is a component of the adaptation-motor complex, mediating mechanoelectrical transduction adaptation in stereocilia.
- MYO1C also localizes to the nucleus, where it associates with transcriptionally active ribosomal genes and facilitates transcription initiation by cooperating with the WICH chromatin-remodeling complex.
- Subcellularly, MYO1C is found in the cytoplasm, nucleus (nucleoplasm, nucleolus), cell cortex, stereocilium membrane, cytoplasmic vesicles, and ruffle membrane.
- The protein is subject to post-translational modifications including acetylation, methylation, and phosphorylation, and contains actin-binding, ATP-binding, and calmodulin-binding domains.
Experimental Guidance and Technical Tips
- The immunogen used spans amino acids 769-1028 of human MYO1C, corresponding to the C-terminal tail domain, which may confer target specificity.
- For Western blot, the predicted molecular weight is 122 kDa; consider including appropriate molecular weight markers and optimizing blocking and antibody incubation conditions.
- In immunohistochemistry (paraffin), antigen retrieval and antibody titration may be necessary for optimal signal in human, mouse, or rat tissues.
- For immunoprecipitation and ELISA, validate the antibody's performance in the relevant sample system and consider using positive and negative controls.
