Biological Background: MYL9 Function and Localization
- Myosin regulatory light polypeptide 9 (MYL9), also known as 20 kDa myosin light chain (LC20), MLC-2C, myosin regulatory light chain MRLC1, is a regulatory subunit of myosin.
- Plays a critical regulatory role in smooth muscle and nonmuscle cell contractile activity via phosphorylation; involved in cytokinesis, receptor capping, and cell locomotion. Related references: PMID:11942626 PMID:2526655
- In myoblasts, may regulate PIEZO1-dependent cortical actomyosin assembly important for myotube formation (by similarity).
- Subcellular localization: Found in the cytoplasm, associated with the cytoskeleton and cell cortex.
- Tissue specificity: Expressed in smooth muscle tissues and in a subset of nonmuscle cells.
- Keywords highlight post-translational modifications including acetylation and phosphorylation, calcium and metal binding, and classification as a motor protein and muscle protein.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 1-100 of human MYL9 (NP_006088.2); antibodies may recognize epitopes within the N-terminal region. For western blot, a band around 20 kDa is expected; optimize lysis and loading controls for your sample type.
- As a polyclonal antibody, batch-to-batch variation is possible; validate performance in your specific assay system with appropriate positive and negative controls.
- For immunofluorescence (IF-P), optimize fixation and permeabilization for cortical or cytoskeletal detection; co-staining with actin or myosin markers can confirm localization.
- The antibody is reported for WB, IF-P, and ELISA and cross-reacts with human, mouse, and rat samples; confirm cross-reactivity experimentally if used in other species.
