Biological Background: MYL9 Function and Localization
- MYL9 encodes myosin regulatory light polypeptide 9, also known as 20 kDa myosin light chain (LC20), MLC-2C, and myosin regulatory light chain 2, smooth muscle isoform.
- As a myosin regulatory subunit, MYL9 plays an important role in regulating both smooth muscle and nonmuscle cell contractile activity via its phosphorylation. It is implicated in cytokinesis, receptor capping, and cell locomotion. Related references: PMID:11942626 PMID:2526655
- In myoblasts, MYL9 may regulate PIEZO1-dependent cortical actomyosin assembly involved in myotube formation (By similarity).
- Subcellularly, the protein localizes to the cytoplasm, cytoskeleton, and cell cortex.
- Tissue expression is prominent in smooth muscle tissues and in some nonmuscle cells.
- UniProt keywords highlight features such as Acetylation, Alternative splicing, Calcium binding, Metal-binding, Phosphoprotein, and Repeat regions, indicating multiple regulatory and functional domains.
Experimental Guidance and Technical Tips
- The immunogen used is an N-terminal peptide (aa 1–100), which may be suitable for detecting total MYL9 protein regardless of phosphorylation state; however, users should validate specificity in their own sample system.
- For western blotting, the predicted molecular weight is ~20 kDa. Slight variations in migration may occur due to post-translational modifications or splice isoforms.
- For ELISA applications, consider pairing with a detection antibody recognizing a non-overlapping epitope to avoid competition; a sandwich ELISA format may be optimized with appropriate positive controls.
- Cross-reactivity is reported for human and mouse; for other species, check sequence homology to the immunogen region and consider testing experimentally.
