Biological Background: MYL2 Function and Localization

  • MYL2 encodes ventricular/cardiac myosin regulatory light chain 2 (MLC-2v), also known as cardiac myosin light chain 2 (MLC-2), a key component of the sarcomeric thick filament in cardiac and slow skeletal muscle.
  • Functions as a contractile protein crucial for heart development; upon phosphorylation, it regulates cross-bridge cycling kinetics, increases myosin lever arm stiffness, and enhances calcium sensitivity of contraction force. Related references: PMID:23365102 PMID:32453731
  • Localizes to the A band of sarcomeres within myofibrils in the cytoplasm of cardiomyocytes.
  • Highly expressed in type I (slow-twitch) muscle fibers.
  • Associated with familial hypertrophic cardiomyopathy (CMH10) and myofibrillar myopathy (MFM12) when mutated.
  • Contains calcium-binding EF-hand motifs and is regulated by phosphorylation and methylation, suggesting metal-dependent regulatory mechanisms.

Experimental Guidance and Technical Tips

  • The immunogen used (recombinant protein covering amino acids 1–166) suggests the antibody recognizes an epitope within the N-terminal region of MYL2, which may detect the full-length ~19 kDa protein in western blotting; a band near this molecular weight is expected in cardiac and slow skeletal muscle lysates.
  • Due to high sequence conservation, cross-reactivity with mouse and rat orthologs is confirmed; consider validation in the relevant species' tissue extracts.
  • For immunofluorescence (IF-P), anticipate sarcomeric A band staining in cardiac muscle sections; appropriate fixation and permeabilization should be optimized.
  • This antibody is applicable in ELISA; consider its use in quantitative assays targeting MYL2 in serum or tissue homogenates, but validate with appropriate controls.
