Biological Background: Myeloperoxidase (MPO) Function and Localization

  • MPO is a heme-containing peroxidase that plays a critical role in the innate immune system, primarily expressed in neutrophils and monocytes.
  • It mediates the formation of neutrophil extracellular traps (NETs) by translocating to the nucleus, binding to nucleosomes, and inducing chromatin decondensation and DNA unwrapping in an ATP-independent manner. Related references: PMID:40963017
  • As a peroxidase, MPO catalyzes the production of hypochlorous acid (HOCl), hypobromous acid, and other reactive oxidants from hydrogen peroxide, which are essential for microbicidal activity against a broad range of pathogens. Related references: PMID:16125131, PMID:17074761, PMID:17438335
  • MPO also oxidizes nitrite to nitrogen dioxide radical, contributing to protein nitration and modulation of inflammatory signaling. Related references: PMID:10777476, PMID:8385644, PMID:9450756
  • The enzyme localizes to azurophilic granules (lysosomes) of neutrophils and is released into the phagosome or extracellularly upon degranulation; it has also been detected in the nucleus and on chromosomes.
  • Post-translational modifications include heme binding, calcium coordination, and N-linked glycosylation; the mature protein is a tetramer of two light chains and two heavy chains derived from a single precursor.
  • MPO can chlorinate and nitrate apolipoprotein A-I (APOA1), impairing cholesterol efflux and potentially contributing to atherosclerosis. Related references: PMID:15326314, PMID:15498770, PMID:15574409
  • The peroxidase activity can be inhibited by the S.aureus protein SPIN, representing an immune evasion strategy.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the C-terminal portion of the MPO heavy chain (amino acids 606–745), which may target a conserved region across species, consistent with reported cross-reactivity with mouse and rat.
  • For Western blotting, the predicted molecular weight of MPO is approximately 84 kDa (precursor) or 59 kDa (heavy chain) and 13.5 kDa (light chain) after processing; consider optimizing sample preparation to detect both precursor and mature forms.
  • In immunohistochemistry (IHC-P) and immunofluorescence (IF-P), validate the antibody in relevant tissue or cell systems, such as neutrophil-rich inflamed tissue or isolated blood cells, to confirm specific staining.
  • Perform appropriate controls (e.g., isotype control, peptide blocking) and titrate antibody concentrations for each application to achieve an optimal signal-to-noise ratio.
