Biological Background: Myeloperoxidase (MPO) Function and Localization

• Myeloperoxidase is an iron-containing heme protein expressed specifically in neutrophils and monocytes. It is synthesized as a precursor and processed into light and heavy chains.
• Functions in neutrophil extracellular trap (NET) formation: it translocates to the nucleus, binds nucleosomes, and drives chromatin decondensation and DNA extrusion, trapping pathogens. Related references: PMID:40963017

• For IHC-P, antigen retrieval may be required; optimize using FFPE sections from tissues rich in neutrophils, e.g., bone marrow or inflamed tissue.
• For IF/ICC, consider permeabilization protocols to access intracellular granular and nuclear MPO; use appropriate blocking to reduce non-specific binding.
