Biological Background: Myeloblastin (Proteinase 3/PR3) Function and Localization
- Protein Identity and Aliases: Myeloblastin, also known as Proteinase 3 (PR3), C-ANCA antigen, Leukocyte proteinase 3, Neutrophil proteinase 4 (NP-4), P29, Wegener autoantigen. It belongs to the serine protease family.
- Proteolytic Activity on Extracellular Matrix: Functions as a serine protease that degrades elastin, fibronectin, laminin, vitronectin, and collagen types I, III, and IV (in vitro). Related references: PMID:2033050, PMID:28240246, PMID:3198760
- Endothelial Barrier Regulation: Cleaves and activates receptor F2RL1/PAR-2, enhancing endothelial cell barrier function and vascular integrity during neutrophil transendothelial migration. Related references: PMID:23202369
- Neutrophil Migration: Plays a role in neutrophil transendothelial migration, probably when associated with CD177. Related references: PMID:22266279
- Inflammatory Signaling: Triggers inflammatory processes in neutrophils by interacting with ADGRG3 upstream of F2RL1/PAR2 activation. Related references: PMID:36302784
- Subcellular Localization: Localizes to cytoplasmic granules, can be secreted, and is also found at the cell membrane and in membrane rafts.
- Tissue Expression: Expressed in polymorphonuclear leukocytes and neutrophils (at protein level). Related references: PMID:2033050, PMID:3198760, PMID:7897245, PMID:17244676, PMID:18462208, PMID:21193407, PMID:22266279, PMID:28240246
- Structural and Biochemical Features: Possesses a signal peptide and is synthesized as a zymogen; contains disulfide bonds and glycosylation sites (glycoprotein). It is a reference proteome member.
Experimental Guidance and Technical Tips
- The immunogen used spans the N-terminal region (amino acids 1–100), which includes the signal peptide and part of the mature enzyme. The antibody is likely to recognize both the proform and the active protease.
- For immunofluorescence on paraffin-embedded sections (IF-P), consider using neutrophil-rich tissue (e.g., inflamed human tissue, bone marrow) or isolated neutrophils. Optimize antigen retrieval conditions.
- In ELISA, this monoclonal antibody can be used as a capture or detection reagent; validate its pairing with other PR3-specific antibodies to maximize sensitivity.
- Since PR3 is secreted upon neutrophil activation, stimulated cell supernatants can serve as a positive control for ELISA or other detection methods.
