- MBP is one of the most abundant protein components of central nervous system (CNS) myelin, together with proteolipid protein (PLP), and plays essential roles in myelin membrane formation and stabilization.
- Alternative splicing produces classic MBP isoforms (isoform 4–14) and non-classic Golli-MBP isoforms (isoform 1–3), each with distinct expression patterns and functions.
- The smaller classic MBP isoforms are implicated in the remyelination of denuded axons in multiple sclerosis.
- Golli-MBP isoforms are expressed in the developing brain and immune system, potentially acting as components of transcriptional complexes and in T-cell signaling pathways.
- MBP induces T-cell proliferation, linking it to autoimmune responses in diseases such as multiple sclerosis.
- Subcellularly, MBP localizes to the myelin membrane and, for some isoforms, the nucleus.

- In immunohistochemistry (IHC-P) and immunofluorescence (IF-P), validate the antibody in appropriate CNS or PNS tissue sections, considering the expected myelin localization.
