Biological Background: MyD88 Function and Localization
- MyD88 (myeloid differentiation primary response protein) is a universal adapter protein in the innate immune system, transducing signals from Toll-like receptors (TLRs) and IL-1 receptors (IL-1R) to activate NF-kappa-B and cytokine production. Related references: PMID:15361868, PMID:18292575, PMID:33718825
- As an adaptor, MyD88 recruits IRAK1 and IRAK2, and interacts with TRAF6 and IRF7, ultimately triggering pro-inflammatory cytokine secretion and immune activation. Related references: PMID:15361868, PMID:19506249, PMID:24316379
- MyD88 directly mediates IL-8 transcriptional upregulation and participates in IL-18 signaling. Related references: PMID:9013863
- In response to viral GU-rich RNA (e.g., from SARS-CoV-2, HIV-1) via TLR8, MyD88 induces NLRP3 inflammasome activation and IL-1β release. Related references: PMID:33718825
- In intestinal epithelial cells, MyD88 signaling is required for gut homeostasis and regulates antimicrobial peptide expression, such as REG3G (By similarity).
- MyD88 localizes to both cytoplasm and nucleus, and its activity is modulated by post-translational modifications including phosphorylation and ubiquitination.
- The MYD88 gene undergoes alternative splicing, producing isoforms that can act as dominant-negative regulators.
- MyD88 is ubiquitously expressed across human tissues.
Experimental Guidance and Technical Tips
- Immunogen design: The antibody was raised against a recombinant fragment spanning amino acids 60–309 of human MyD88, covering the TIR domain critical for dimerization and signaling. Consider this when interpreting functional assay results.
- Western blotting: A specific band is expected at ~33 kDa. Additional bands may correspond to splice variants or modified forms; validate with appropriate controls.
- IHC-P: Heat-induced epitope retrieval may optimize staining. Validate in positive and negative tissue controls.
- ELISA: The polyclonal nature may provide broad epitope recognition; evaluate for sandwich ELISA by pairing with a monoclonal capture antibody if needed.
- Cross-reactivity: Detects endogenous MyD88 in human, mouse, and rat samples per specification. Verify species-specific expression in your model system.
