Biological Background: MVK Function and Localization
- Mevalonate kinase (MVK) is an essential enzyme in the mevalonate pathway, catalyzing the ATP-dependent phosphorylation of mevalonate to form mevalonate 5-phosphate, a critical step in cholesterol and isoprenoid biosynthesis. Related references: PMID:11278915 PMID:18302342 PMID:9325256
- The enzyme belongs to the galactokinase family and requires magnesium ions for its kinase activity.
- MVK is primarily localized in the cytoplasm and peroxisomes, consistent with its role in early steps of cholesterol biosynthesis.
- Genetic mutations in MVK are associated with human diseases, including mevalonate kinase deficiency (MKD) which manifests as hyper-IgD syndrome and mevalonic aciduria, as well as porokeratosis (POROK3).
