Biological Background: Munc 13-1 Function and Localization
- Protein unc-13 homolog A (UNC13A, also known as Munc13-1) is a 193 kDa multi-domain protein containing C2 domains, phorbol ester/diacylglycerol-binding zinc fingers, and a C-terminal MUN domain essential for synaptic vesicle priming.
- It functions as a priming factor in synaptic vesicle exocytosis, essential for most excitatory/glutamatergic synapses, and participates in activity-dependent refilling of the readily releasable vesicle pool (RRP) (By similarity).
- Acts downstream of diacylglycerol second messenger signaling to mature vesicles for fusion, linking intracellular calcium signals to regulated neurotransmitter and hormone secretion.
- Expressed in pancreatic islet cells, Munc13-1 primes insulin secretory granules for release, contributing to glucose-stimulated insulin secretion.

PMID:23999003 - Localizes to the presynaptic active zone, cytoplasm, and cell membrane, where it positions synaptic vesicles near calcium channels for rapid exocytosis.
- Contains metal-binding domains (calcium, zinc) and phorbol-ester/DAG-type zinc fingers, reflecting its regulation by second messengers and metal ions.
- Mutations in UNC13A are implicated in autosomal dominant epilepsy and intellectual disability, underscoring its critical role in synaptic transmission and neurodevelopment.
Experimental Guidance and Technical Tips
- The immunogen is recombinant human Munc13-1; for species not listed under Cross Reactivity, sequence homology should be checked to predict potential cross-reactivity.
- For Western blot, expect a band near 193 kDa; use low-percentage gels and extended transfer times for high molecular weight proteins, and consider reducing conditions.
- For ELISA, validate detection using recombinant protein or lysates from tissues expressing Munc13-1 (e.g., brain, pancreas).
- Include appropriate negative controls (e.g., knockout cell lysates) to confirm antibody specificity.
