  • Multimerin-1 (MMRN1), also known as EMILIN-4 or elastin microfibril interfacer 4, is a secreted glycoprotein belonging to the EMILIN family of extracellular matrix proteins.
  • It functions as a carrier protein for platelet factor V/Va, playing a critical role in the storage and stabilization of factor V within platelet alpha granules.
  • Upon platelet activation, MMRN1 is released and may limit factor Va-dependent thrombin generation, thereby regulating coagulation.
  • MMRN1 serves as a ligand for integrin alpha-IIb/beta-3 and integrin alpha-V/beta-3 on activated platelets, suggesting a role as an adhesive protein in platelet plug formation and thrombus stabilization.
  • Subcellularly, MMRN1 is secreted and stored in platelet alpha granules and endothelial cell Weibel-Palade bodies, from which it is released upon cellular activation and deposited onto the endothelium and subendothelium.
  • Synthesized primarily by endothelial cells and megakaryocytes, MMRN1 is found in vascular tissues such as placenta, lung, and liver, but is not detected in normal plasma.
  • The protein contains an N-terminal signal peptide, coiled-coil domains, EGF-like domains, and disulfide bonds, which contribute to its structural stability and multimerization.
