Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide from the central region (aa 1550–1900) of human Mucin 5AC. This region may be glycosylated in the native protein; verify that the antibody recognizes the mature glycoprotein under your experimental conditions.
- For Western blot, consider using reducing agents to break disulfide bonds, as the native mucin forms very large multimers (>500 kDa). Migration may be affected by glycosylation, so use appropriate molecular weight markers and optimize transfer conditions.
- This polyclonal antibody cross-reacts with mouse and rat Mucin 5AC, making it suitable for translational studies in rodent models. Validate species reactivity in your specific tissue lysates.
- ELISA applications may require coating conditions (e.g., carbonate buffer, pH 9.6) and blocking steps optimized for large glycoproteins. Include a negative control such as a non-mucin protein.
