  • MUC4 regulates cellular behavior through anti-adhesive effects on cell-cell and cell-extracellular matrix interactions and serves as an intramembrane ligand for ERBB2, inducing specific ERBB2 phosphorylation to promote proliferation and differentiation of epithelial cells.
  • Interaction with ERBB2 enhances expression of CDKN1B; formation of a MUC4-ERBB2-ERBB3-NRG1 complex down-regulates CDKN1B, leading to repression of apoptosis and stimulation of proliferation, which may contribute to tumor growth.
  • Alternative splicing yields membrane-bound and secreted isoforms; the protein localizes to the cell membrane and is also secreted.
  • It is expressed in a wide range of epithelial tissues (thymus, thyroid, lung, trachea, esophagus, stomach, small intestine, colon, testis, prostate, ovary, uterus, placenta, mammary and salivary glands) and in carcinomas of lung, upper aerodigestive tract, breast, biliary tract, colon, and cervix; minimal expression in normal pancreas but high expression in pancreatic tumors.
