- MUC4 is a high-molecular-weight (542 kDa) transmembrane glycoprotein belonging to the mucin family, characterized by extensive O-glycosylation and a large extracellular domain with EGF-like repeats.

- MUC4 functions as an intramembrane ligand for the receptor tyrosine kinase ERBB2, inducing ERBB2 phosphorylation and modulating downstream signaling pathways to influence cell proliferation, differentiation, and apoptosis; it can form complexes with ERBB3 and NRG1 that repress apoptosis and promote tumor growth. - Given the large size of MUC4 (542 kDa), consider using low percentage SDS-PAGE (e.g., 4-6%) for western blot to enhance transfer efficiency. Extended transfer times and low voltage may improve detection.
- For IHC-P, antigen retrieval methods (such as heat-induced epitope retrieval in citrate buffer, pH 6.0) may be necessary; validation in known positive and negative tissue controls (e.g., pancreatic tumor vs. normal pancreas) is recommended.
- Cross-reactivity with rat has been reported; validation in rat samples should be performed to confirm suitability for rodent studies.
