Biological Background: Mucin-1 Function and Localization

  • Mucin-1 (MUC1) is a large, heavily glycosylated transmembrane protein also known as CA15-3, EMA, and KL-6.
  • The alpha subunit mediates cell adhesion and can act as both adhesion and anti-adhesion molecule, providing a protective barrier on epithelial surfaces.
  • The beta subunit participates in intracellular signaling through phosphorylation and protein interactions, modulating ERK, SRC, and NF-kappa-B pathways.
  • MUC1 is expressed on the apical surface of epithelial cells in tissues including airway, breast, and uterus, and is also found in activated T-cells.
  • Overexpression of MUC1, particularly isoform Y, is associated with epithelial tumors such as breast and ovarian cancers.
  • Subcellularly, MUC1 localizes to the apical cell membrane, but is also secreted and found in cytoplasm and nucleus.

  • Predicted molecular weight is ~122 kDa, but heavy glycosylation may cause smearing or higher apparent mass; validate with deglycosylation controls if needed.
  • For IHC-P, antigen retrieval may be necessary; validate signal in both normal and tumor tissues to confirm specificity.
  • In ELISA, use purified or recombinant MUC1 as coating antigen; note that MUC1 is secreted and can be detected in cell supernatants or biological fluids.
  • The antibody cross-reacts with mouse and rat MUC1, enabling use in animal models; include appropriate negative controls to confirm species cross-reactivity.
