Biological Background: MTMR9 Function and Localization

  • MTMR9 is an inactive myotubularin-related phosphatase adapter, encoded by the MTMR9 gene (also known as C8orf9 or MTMR8), with multiple isoforms produced by alternative splicing.
  • Acts as an adapter for the lipid phosphatases MTMR6, MTMR7, and MTMR8, enhancing their catalytic activities. Related references: PMID:19038970 PMID:22647598
  • Specifically increases MTMR6 activity toward phosphatidylinositol 3,5-bisphosphate and its binding to phosphorylated phosphatidylinositols, while also stabilizing MTMR6 protein levels through complex formation. Related references: PMID:19038970 PMID:22647598
  • Increases MTMR8 catalytic activity toward phosphatidylinositol 3-phosphate and stabilizes MTMR8 protein levels. Related references: PMID:22647598
  • Plays a role in the late stages of macropinocytosis, likely by regulating MTMR6-mediated dephosphorylation of phosphatidylinositol 3-phosphate at membrane ruffles. Related references: PMID:24591580
  • Negatively regulates autophagy through its association with MTMR8, and suppresses DNA damage-induced apoptosis through its interaction with MTMR6. Related references: PMID:22647598 PMID:19038970
  • Does not bind mono-, di-, and tri-phosphorylated phosphatidylinositols, phosphatidic acid, or phosphatidylserine. Related references: PMID:19038970
  • Localizes to the cytoplasm, perinuclear region, endoplasmic reticulum, and cell projections including ruffle membranes.

Experimental Guidance and Technical Tips

  • The immunogen comprises the N-terminal 250 amino acids, which may recognize multiple isoforms; verify specificity in your experimental system.
  • For western blot, expect a predominant band at approximately 63 kDa; consider using appropriate positive and negative controls.
  • Validate antibody reactivity in human, mouse, and rat samples; further dilution optimization may be needed for IHC-P and IF/ICC.
  • For immunofluorescence applications, mild fixation and permeabilization conditions are recommended to preserve both cytosolic and membrane-associated MTMR9 pools.
