Biological Background: MTMR6 Function and Localization

  • MTMR6 (Myotubularin-related protein 6) is a lipid phosphatase that specifically dephosphorylates phosphatidylinositol 3-phosphate (PtdIns(3)P) and phosphatidylinositol 3,5-bisphosphate (PtdIns(3,5)P2) at the D-3 position, and binds with high affinity to these substrates. Related references: PMID:19038970 PMID:22647598
  • It negatively regulates ER-Golgi protein transport.
  • In association with MTMR9, MTMR6 participates in the late stages of macropinocytosis by dephosphorylating PtdIns(3)P in membrane ruffles. Related references: PMID:24591580
  • MTMR6 acts as a negative regulator of the KCNN4/KCa3.1 potassium channel activity in CD4+ T-cells, likely by reducing intracellular levels of PtdIns(3)P. Related references: PMID:15831468
  • It negatively regulates proliferation of reactivated CD4+ T-cells. Related references: PMID:16847315
  • The formation of the MTMR6-MTMR9 complex stabilizes both protein levels and negatively regulates DNA damage-induced apoptosis. Related references: PMID:19038970 PMID:22647598
  • Subcellular localization includes the cytoplasm, endoplasmic reticulum, ER-Golgi intermediate compartment, ruffle membrane, and perinuclear region.
  • MTMR6 is expressed in CD4+ T-cells and is subject to alternative splicing and phosphorylation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to amino acids 382-621 of human MTMR6, a region expected to be present in both full-length and alternatively spliced isoforms; consider validating in relevant cell lines.
  • For Western blot, the predicted molecular weight is ~72 kDa; use whole-cell lysates from human, mouse, or rat T-cells or tissues expressing MTMR6.
  • ELISA applications may be optimized for detecting endogenous or recombinant MTMR6; include appropriate positive and negative controls.
  • Because MTMR6 localizes to membrane ruffles and intracellular compartments, consider fractionation or enrichment strategies to improve detection sensitivity in WB.
  • Cross-reactivity with mouse and rat has been reported, but validate the antibody in your specific experimental model before large-scale studies.
