Biological Background: MTMR4 Function and Localization
- Encodes the phosphatidylinositol-3,5-bisphosphate 3-phosphatase MTMR4, also known as FYVE-DSP2 or ZFYVE11, a member of the myotubularin-related protein family.
- Catalyzes the removal of the D-3 phosphate from phosphatidylinositol 3-phosphate (PI3P) and phosphatidylinositol 3,5-bisphosphate, generating phosphatidylinositol and phosphatidylinositol 5-phosphate, respectively. Related references: PMID:11302699 PMID:16787938 PMID:20736309
- Reduces PI3P levels on a subset of endosomal membranes, negatively regulating endocytic recycling and the trafficking/maturation of endosomes toward lysosomes. Related references: PMID:16787938 PMID:20736309 PMID:29962048
- Controls phagocytosis and phagosome maturation via PI3P turnover on phagosome membranes. Related references: PMID:31543504
- Modulates innate immune responses by regulating PI3P levels in immune cells. Related references: PMID:30944173
- Acts as a molecular adapter during mitotic cytokinesis to regulate midbody abscission. Related references: PMID:25659891
- Negatively regulates TGF-beta and BMP signaling by promoting Smad protein dephosphorylation and retention in endosomes. Related references: PMID:20061380 PMID:23150675
- Localizes predominantly to membranes of early, recycling, and late endosomes, as well as to phagosome membranes.
- Widely expressed, with high levels in brain, heart, kidney, spleen, liver, colon, testis, muscle, placenta, thyroid gland, pancreas, ovary, prostate, skin, peripheral blood, and bone marrow.
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 570–820 of human MTMR4; the resulting polyclonal antibody may recognize epitopes within this C‑terminal region.
- For IHC‑P, consider performing antigen retrieval and titrating the antibody to determine the optimal working dilution.
- In ELISA, use the antibody as a detection or capture reagent after validating performance with your specific antigen and assay conditions.
- Cross‑reactivity with mouse is stated; if working with other species, validate in the relevant sample system.
- Because it is a polyclonal preparation, lot‑to‑lot consistency should be verified in critical applications.
