Biological Background: MTH1 Function and Localization
- Oxidized purine nucleoside triphosphate hydrolase, also known as MTH1, NUDT1, 8-oxo-dGTPase, and Nucleoside diphosphate-linked moiety X motif 1, is a sanitizer of the oxidized nucleotide pool. Related references: PMID:10608900, PMID:12857738, PMID:22556419
- Catalyzes hydrolysis of 2-oxo-dATP, 2-oxo-ATP, 8-oxo-dGTP, and 8-oxo-dATP to prevent incorporation into DNA and subsequent A:T to C:G and G:C to T:A transversions. Related references: PMID:10373420, PMID:11139615, PMID:10608900, PMID:11756418
- Also hydrolyzes methylated purine nucleoside triphosphates, providing antimutagenic activity that protects cells from oxidative stress. Related references: PMID:30304478, PMID:32144205, PMID:10608900, PMID:24695224
- Belongs to the Nudix hydrolase family, containing a Nudix motif 1; requires magnesium as a cofactor and may bind RNA.
- Localizes to the cytoplasm, cytosol, mitochondrial matrix, and nucleus; widely expressed with highest levels in thymus, testis, embryo, and proliferating blood lymphocytes.
- Molecular weight is approximately 18 kDa (UniProt: 17952 Da); alternative initiation generates isoforms.
Experimental Guidance and Technical Tips
- The immunogen used is a recombinant fusion protein covering amino acids 50–200 of human MTH1 (P36639), which may be suitable for detecting both cytoplasmic and mitochondrial pools of the enzyme.
- For western blot (WB), the predicted molecular weight is approximately 18 kDa; consider using a reducing gel and loading appropriate controls from human cell lysates known to express MTH1 (e.g., thymus, testis).
- For ELISA, validate the antibody’s performance with recombinant MTH1 protein or native protein in human sample matrices.
- Since MTH1 is widely expressed, background may vary; optimize blocking and antibody dilution for each sample type.
