Biological Background: MTH1 Function and Localization
- MTH1, also known as NUDT1 (Nudix motif 1), is a sanitizer of the oxidized nucleotide pool, hydrolyzing oxidized purine nucleoside triphosphates to prevent DNA damage. Related references: PMID:10608900, PMID:12857738, PMID:22556419
- It catalyzes the hydrolysis of damaged nucleotides such as 2-oxo-dATP, 2-oxo-ATP, 8-oxo-dGTP, and 8-oxo-dATP, thereby preventing their misincorporation into DNA. Related references: PMID:10373420, PMID:11139615
- Through this activity, MTH1 prevents A:T to C:G and G:C to T:A transversions, acting as a critical antimutagenic enzyme in the defense against oxidative DNA damage. Related references: PMID:10373420, PMID:10608900, PMID:11756418
- MTH1 also hydrolyzes methylated purine nucleoside triphosphates, further protecting the genome from alkylation-induced mutations. Related references: PMID:30304478, PMID:32144205
- The enzyme is localized in the cytoplasm, cytosol, mitochondrial matrix, and nucleus, aligning with its role in safeguarding both nuclear and mitochondrial genomes.
- MTH1 is widely expressed, with the highest levels found in thymus, testis, embryo, and proliferating blood lymphocytes, indicating importance in tissues with high replication rates.
- The protein possesses a Nudix hydrolase domain, requires magnesium for activity, and has a molecular weight of approximately 18 kDa. Three-dimensional structures are available, and alternative initiation variants exist.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 1-179 of human MTH1, allowing recognition of the full-length protein. For Western blot, a band of approximately 18 kDa is expected; validate with a positive control lysate from human cell lines.
- For ELISA, consider using the recombinant MTH1 protein as a coating antigen and testing a dilution series of the antibody to establish optimal working concentrations.
- This antibody is reported to react with human MTH1; cross-reactivity with other species has not been tested. If using in other species, verify sequence homology and validate experimentally.
