Biological Background: MTERFD1 Function and Localization
- MTERFD1 (also known as mTERF3) is a mitochondrial transcription termination factor encoded by the MTERF3 gene.
- Binds promoter DNA and regulates initiation of mitochondrial transcription. Related references: PMID:17662942
- Required for normal mitochondrial transcription, translation, and assembly of respiratory complexes (by similarity).
- Maintains 16S rRNA levels and functions in mitochondrial ribosome assembly by regulating biogenesis of the 39S ribosomal subunit (by similarity).
- Alternative splicing produces multiple protein isoforms.
- Localizes to the mitochondrion.
- Highly expressed in heart, liver, kidney, and testis; lower expression in brain, spleen, and lung.
- Contains a DNA-binding domain and a transit peptide for mitochondrial import.
Experimental Guidance and Technical Tips
- For Western blot, consider using mitochondrial-enriched lysates to enhance detection of MTERFD1, given its mitochondrial localization.
- Validate the antibody in human and mouse sample systems to confirm cross-reactivity and specificity.
- Optimize antibody dilution for each application (WB/ELISA) by performing a titration series.
- The immunogen spans amino acids 69–300, which excludes the N-terminal transit peptide; this may allow detection of both precursor and mature protein forms.
- Confirm band size at approximately 48 kDa; additional bands may represent isoforms from alternative splicing.
