- MTDH (Metadherin, also known as Protein LYRIC, AEG-1, 3D3/LYRIC) is a multifunctional protein encoded by the MTDH gene.
- It functions as a transcriptional regulator: down-regulating SLC1A2/EAAT2 promoter activity and activating NF-kappa-B transcription factor, promoting oncogenic signaling.
- Promotes anchorage-independent growth, a hallmark of cellular transformation, and enhances metastasis to lung, bone, and brain by facilitating tumor cell seeding to target organ endothelia.
- Induces chemoresistance, contributing to treatment failure in various cancers.
- Localizes to endoplasmic reticulum and nuclear membranes, tight junctions, nucleolus, and perinuclear cytoplasm, suggesting roles in cell adhesion and nuclear function.
- Widely expressed in normal tissues with highest levels in muscle-dominant organs and endocrine glands; significantly overexpressed in multiple cancer types, including breast, brain, prostate, and melanoma.
