Biological Background: MTDH Function and Localization

  • MTDH (Metadherin, also known as Protein LYRIC, AEG-1, 3D3/LYRIC) is a multifunctional protein encoded by the MTDH gene.
  • It functions as a transcriptional regulator: down-regulating SLC1A2/EAAT2 promoter activity and activating NF-kappa-B transcription factor, promoting oncogenic signaling.
  • Promotes anchorage-independent growth, a hallmark of cellular transformation, and enhances metastasis to lung, bone, and brain by facilitating tumor cell seeding to target organ endothelia.
  • Induces chemoresistance, contributing to treatment failure in various cancers.
  • Localizes to endoplasmic reticulum and nuclear membranes, tight junctions, nucleolus, and perinuclear cytoplasm, suggesting roles in cell adhesion and nuclear function.
  • Widely expressed in normal tissues with highest levels in muscle-dominant organs and endocrine glands; significantly overexpressed in multiple cancer types, including breast, brain, prostate, and melanoma.
  • Post-translationally modified by phosphorylation and acetylation; contains a single transmembrane helix.

Experimental Guidance and Technical Tips

  • The immunogen covers the N-terminal region (aa 1-260) of human MTDH; when designing isoform-specific experiments, verify sequence conservation across isoforms.
  • For WB, the predicted molecular weight is ~64 kDa, but post-translational modifications may shift the observed band; use appropriate molecular weight markers and loading controls.
  • This polyclonal antibody is suitable for ELISA; consider using paired monoclonal/polyclonal antibodies for sandwich ELISA development.
  • For IHC-P and IF/ICC, optimize antigen retrieval methods (e.g., heat-induced epitope retrieval) and permeabilization (e.g., Triton X-100) based on the target's membrane/organelle localization.
  • Cross-reactivity with mouse and rat MTDH is listed; validate specificity in your model system by comparing with known positive and negative controls.
