Biological Background: MTA3 Function and Localization

  • MTA3 (Metastasis-associated protein MTA3) is a component of the nucleosome remodeling and deacetylase (NuRD) complex, which plays a critical role in chromatin remodeling and transcriptional regulation. Related references: PMID:12705869, PMID:16428440, PMID:28977666
  • It maintains normal epithelial architecture by repressing SNAI1 transcription in a histone deacetylase-dependent manner, thereby regulating E-cadherin levels. Related references: PMID:12705869
  • MTA3 also contributes to transcriptional repression mediated by BCL6, a key regulator in germinal center formation. Related references: PMID:15454082
  • The protein is predominantly localized in the nucleus, but can also be found in the cytoplasm.
  • Tissue expression is restricted to germinal centers of lymphoid tissues, with no detection in nonepithelial cells.
  • MTA3 contains zinc-finger domains, suggesting DNA-binding capability, and is subject to phosphorylation.
  • Alternative splicing produces multiple isoforms, adding to functional diversity.

Experimental Guidance and Technical Tips

  • The immunogen spans amino acids 40–330, covering multiple domains of MTA3; this polyclonal antibody may recognize various isoforms and denatured epitopes, making it suitable for WB.
  • For Western blot, a band around 68 kDa is expected; however, due to post-translational modifications and isoform variation, verify the observed molecular weight in your sample system.
  • Given the cross-reactivity with mouse, this antibody can be considered for murine models, but we recommend validating the expression pattern in your specific tissue or cell line.
  • For ELISA, optimize coating conditions and detection system, as polyclonal antibodies may require careful titration to minimize background.
  • Note that MTA3 is predominantly nuclear; appropriate lysis and fractionation protocols may enhance detection sensitivity.
