Biological Background: MT-ND5 Function and Localization
- MT-ND5 (NADH dehydrogenase subunit 5) is a core subunit of mitochondrial respiratory chain Complex I, catalyzing electron transfer from NADH to ubiquinone, which is essential for the catalytic activity and assembly of complex I. Related references: PMID:15250827
- The protein is encoded by the mitochondrial gene MT-ND5 (aliases: MTND5, NADH5, ND5) and is a component of the inner mitochondrial membrane.
- It contains multiple transmembrane helices, anchoring it within the mitochondrial inner membrane (subcellular location: mitochondrion inner membrane).
- Disease-associated mutations in MT-ND5 are linked to mitochondrial disorders, including Leber hereditary optic neuropathy, Leigh syndrome, and MELAS syndrome.
- Keywords: Electron transport, NAD, Primary mitochondrial disease, Respiratory chain, Translocase, Transport, Ubiquinone, 3D-structure, Proteomics identification.
Experimental Guidance and Technical Tips
- For Western blot, the expected molecular weight is approximately 67 kDa. Consider mitochondrial enrichment for optimal detection.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF-P), permeabilization is recommended due to the mitochondrial membrane localization.
- Validate antibody performance in your specific tissue or cell type, and include appropriate positive and negative controls.
- The immunogen corresponds to a synthetic peptide from mouse MT-ND5 (aa 500-607); antibody reactivity should be tested in the species and application of interest.
