Biological Background: MT-ND2 Function and Localization
- Encodes NADH-ubiquinone oxidoreductase chain 2 (ND2), a core subunit of mitochondrial respiratory chain Complex I, also known as NADH dehydrogenase subunit 2.
- Essential for the catalytic activity and structural assembly of Complex I, facilitating electron transfer from NADH to ubiquinone (PubMed:16996290). Related references: PMID:16996290
- Localizes to the mitochondrial inner membrane, spanning the membrane with multiple transmembrane helices.
- Participates in oxidative phosphorylation, coupling electron transport to proton pumping across the membrane.
- Mutations in MT-ND2 are associated with mitochondrial diseases, including Leber hereditary optic neuropathy (LHON), Leigh syndrome, and Alzheimer disease.
- Classified under keywords: Alzheimer disease, Amyloidosis, Disease variant, Electron transport, Leber hereditary optic neuropathy, Leigh syndrome, Membrane, Mitochondrion, Mitochondrion inner membrane, NAD, Neurodegeneration, Primary mitochondrial disease, Reference proteome, Respiratory chain, Translocase, Transmembrane, Transmembrane helix, Transport, Ubiquinone.
- Identified in proteomics studies, confirming its expression in human tissues.
Experimental Guidance and Technical Tips
- The immunogen is a synthetic peptide spanning amino acids 248-347 of human MT-ND2. Sequence alignment shows high conservation across human, mouse, and rat, aligning with the reported cross-reactivity data. Consider verifying sequence identity for other species before use.
- For Western blot analysis, the predicted molecular weight is approximately 39 kDa. A band near this size is expected in mitochondrial-enriched lysates. Validate with appropriate positive and negative controls.
- In immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), mitochondrial localization should be evident as punctate cytoplasmic staining. Co-staining with a mitochondrial marker is advisable to confirm specificity.
- The antibody is also suitable for ELISA applications. For quantitative assays, pair with a matching detection antibody or use in a sandwich ELISA format.
