Biological Background: MST1/STK4 Function and Localization
- Serine/threonine-protein kinase 4, also known as MST1, KRS2, YSK3, belongs to the STE20-like kinase family.
- Core component of the Hippo signaling pathway, regulating organ size and tumor suppression by phosphorylating LATS1/2, leading to YAP1/WWTR1 inactivation.
- Functions as a stress-activated, pro-apoptotic kinase; caspase-cleavage induces nuclear translocation, chromatin condensation, and DNA fragmentation.
- Phosphorylates histone H2B at Ser14 during apoptosis, and phosphorylates FOXO3 upon oxidative stress, promoting nuclear localization and cell death.
- Inhibits SIRT1-mediated p53 deacetylation, enhancing p53-dependent apoptosis; also acts as an inhibitor of PKB/AKT1.
- Localizes to both cytoplasm and nucleus.
- Ubiquitously expressed, with increased levels observed in prostate cancer.
Experimental Guidance and Technical Tips
- The immunogen is a recombinant fragment spanning the C-terminal region (aa 303–487). The polyclonal antibody may detect full-length MST1/STK4 and caspase-cleaved fragments; consider validating in appropriate apoptotic models.
- For WB, the predicted molecular weight is ~56 kDa. A band near this size should be observed; additional bands may arise from cleavage or post-translational modifications.
- The antibody is confirmed for ELISA. When developing sandwich ELISA assays, consider pairing with a capture antibody that recognizes a different epitope to ensure specificity.
- Cross-reactivity is reported for human, mouse, and rat; always verify performance in the relevant sample matrix and include proper controls.
