Biological Background: MSRB2 Function and Localization
- Encoded by the MSRB2 gene, also known as CBS1 or PILB, the methionine-R-sulfoxide reductase B2 (MSRB2) protein is a mitochondrial member of the methionine sulfoxide reductase family.
- Functions as a stereospecific oxidoreductase that catalyzes the reduction of methionine (R)-sulfoxide back to methionine, repairing oxidatively damaged proteins and maintaining protein function.
- Localizes predominantly to the mitochondria, consistent with its role in protecting mitochondrial proteins from oxidative damage.
- Upon oxidative stress, MSRB2 helps preserve mitochondrial integrity by scavenging reactive oxygen species (ROS), thereby promoting cell survival and protein maintenance.
- Exhibits ubiquitous tissue expression, detected in retina, skeletal muscle, heart, colon, bone marrow, cerebellum, and other tissues, with highest levels in metabolically active organs.
- Contains a transit peptide for mitochondrial targeting and binds zinc as a cofactor, essential for its enzymatic activity.

- The immunogen spans amino acids 10–182 of human MSRB2, covering a large portion of the protein; cross-reactivity with mouse and rat is expected due to sequence conservation.
- For Western blotting, a band around 20 kDa is anticipated, corresponding to the predicted molecular weight of the mature mitochondrial protein.
- Immunohistochemistry applications may reveal a mitochondrial staining pattern; include appropriate controls and validate in relevant tissue sections.
