Biological Background: MSRA Function and Localization
- MSRA (Mitochondrial peptide methionine sulfoxide reductase), also known as PMSR, is a repair enzyme that catalyzes the reversible reduction of methionine sulfoxide residues in proteins back to methionine, restoring protein function damaged by oxidative stress. Related references: PMID:10452521, PMID:12039877
- It plays a crucial role in maintaining lens transparency by repairing oxidized methionine in alpha-crystallin (CRYAA/CRYAB) and cytochrome c (CYCS) (By similarity).
- The enzyme is widely expressed, with highest levels in adult kidney and cerebellum, and moderate in liver, heart ventricles, bone marrow, and hippocampus.
- MSRA localizes to multiple subcellular compartments including the mitochondrial matrix, cytoplasm, nucleus, and membrane, consistent with its role in repairing proteins across organelles.
- The protein undergoes alternative splicing, alternative promoter usage, and alternative translation initiation, generating multiple isoforms with distinct N-termini and targeting signals.
- MSRA contains a redox-active cysteine center and forms a transient disulfide bond during catalysis. It is acetylated and myristoylated, modifications that may regulate its activity or localization.

Experimental Guidance and Technical Tips
- The immunogen used was a recombinant fusion protein spanning amino acids 24–235 of human MSRA (NP_036463.1), covering the mature catalytic domain of the protein. Consider this region for antibody specificity validation.
- For Western blot (WB), the predicted molecular weight is 26 kDa. Because MSRA has alternative isoforms and localizes to multiple compartments, validate band specificity using relevant knockout or knockdown controls.
- For immunohistochemistry (IHC-P) and immunofluorescence (IF/ICC), prefixation/permeabilization conditions should be optimized for the target’s presence in membrane, mitochondrial, and nuclear fractions. Use appropriate organelle markers to confirm subcellular distribution.
- ELISA may be used for quantitative detection; ensure the coating antigen matches the immunogen sequence for optimal sensitivity.
- Cross-reactivity is validated for human, mouse, and rat; for other species, check sequence homology before use.
