  • MSH6, also known as GTBP or p160, is the 160 kDa subunit of the MutS alpha mismatch repair complex.
  • Partners with MSH2 to form MutS alpha, which initiates post-replicative DNA mismatch repair (MMR) by binding single base mismatches and small insertion-deletion loops.
  • Upon DNA mismatch recognition, the MutS alpha complex bends DNA and undergoes an ATP-dependent conformational switch to become a sliding clamp, enabling downstream repair processes.
  • Recruited to chromatin in G1 and early S phase via its PWWP domain binding to trimethylated histone H3 Lys36 (H3K36me3), ensuring early MMR at replication forks.
  • Also implicated in DNA homologous recombination repair.
  • Localizes to the nucleus and chromosomes.
  • Post-translationally modified by acetylation and phosphorylation; mutations in MSH6 are associated with Lynch syndrome (hereditary nonpolyposis colorectal cancer, HNPCC).
