  • MSH6 encodes DNA mismatch repair protein Msh6, also known as GTBP, GTMBP, MutS protein homolog 6, and p160, a key component of the post-replicative mismatch repair (MMR) system.
  • It heterodimerizes with MSH2 to form the MutS alpha complex, which recognizes single-base mismatches and small insertion–deletion loops, initiating DNA repair.
  • MutS alpha binds mismatched DNA, bends the helix, shields approximately 20 base pairs, and recruits MutL alpha to direct downstream repair events including strand discrimination, excision, and resynthesis.
  • ATP binding and hydrolysis by MSH6 act as a molecular switch: mismatched DNA triggers ADP→ATP exchange, inducing a conformational change that converts MutS alpha into a sliding clamp for diffusion along DNA.
  • MSH6’s PWWP domain specifically binds trimethylated lysine 36 of histone H3 (H3K36me3), mediating its recruitment to chromatin in G1 and early S phase to link DNA replication with mismatch recognition.
  • Localized in the nucleus and on chromosomes, MSH6 is critical for maintaining genomic stability; its dysfunction is associated with Lynch syndrome (hereditary nonpolyposis colorectal cancer) and other malignancies.
  • The protein has a molecular weight of approximately 153 kDa, consistent with its large 152 786 Da predicted mass, and undergoes post-translational modifications including acetylation and phosphorylation.

Experimental Guidance and Technical Tips

  • The immunogen corresponds to the N-terminal region (amino acids 1–100) of human MSH6, which contains the PWWP domain; consider that the epitope may be masked by H3K36me3 interactions in fixed chromatin.
  • For Western blot, a band around 153 kDa is expected; due to the large size, use low-percentage SDS-PAGE gels and extended transfer times.
  • In IF/ICC, expect nuclear staining; co-localization with chromatin markers may require permeabilization optimization.
  • Confirm cross-reactivity with mouse and rat samples (human, mouse, rat validated), but verify sequence homology in the immunogen region for other species before use.
