Biological Background: MS4A7 Function and Localization
- MS4A7 (Membrane-spanning 4-domains subfamily A member 7), also known as CD20 antigen-like 4 (CD20L4), CD20/FC-epsilon-RI-beta family member 4 (CFFM4), and four-span transmembrane protein 2 (4SPAN2), is a member of the MS4A family of four-transmembrane proteins.
- MS4A7 is predicted to function as a component of a multimeric receptor complex involved in signal transduction, possibly playing a role in immune cell signaling.
- The protein localizes to the cell membrane and contains four transmembrane helices, characteristic of the MS4A family.
- MS4A7 exhibits ubiquitous expression in normal tissues, with notably higher levels in adult liver, lung, spleen, and heart compared to fetal counterparts, and in non-cancerous versus cancerous tissues, suggesting a role in tissue-specific regulation.
- In the monocytic lineage, MS4A7 expression is low in promonocytes but becomes highly upregulated in mature monocytes, pointing to involvement in monocyte differentiation or function.
- The gene undergoes alternative splicing, and the protein has been identified through proteomics studies, with a molecular weight of approximately 26 kDa.
Experimental Guidance and Technical Tips
- This antibody was raised against a synthetic peptide corresponding to amino acids 100-200 of human MS4A7 (isoform NP_067024.1). The immunogen sequence lies within a region predicted to be intracellular or extracellular; for precise epitope mapping, additional experiments would be required.
- For Western blot (WB), a band around 26 kDa is expected based on the predicted molecular weight; however, post-translational modifications or alternative splicing may alter the detected size.
- In ELISA applications, the antibody may be used for direct or indirect detection of MS4A7 in various sample types; optimization of coating and detection conditions is recommended.
- Cross-reactivity with mouse MS4A7 is reported, making this antibody suitable for comparative studies; however, cross-reactivity with other MS4A family members cannot be excluded due to sequence homology.
