- MRPL27 is a constituent of the large (39S) subunit of the mitochondrial ribosome, essential for mitochondrial protein synthesis.
- It is encoded by the nuclear gene MRPL27 (GeneID: 51264) and imported into mitochondria, where it assembles into the mitoribosome.
- The protein belongs to the bL27m ribosomal protein family, sharing structural homology with bacterial ribosomal protein L27.
- MRPL27 localizes specifically to the mitochondrion, as indicated by its mitochondrial transit peptide and subcellular fractionation data.
- Post-translational removal of the transit peptide yields the mature 16 kDa protein.
- Structural studies have resolved the 3D architecture of the human mitoribosome, including MRPL27 within the large subunit.
Experimental Guidance and Technical Tips
- The immunogen used is an internal peptide (aa 49-148) of human MRPL27; cross-reactivity with human and mouse is expected. For other species, check sequence alignment.
- For Western blotting, the expected molecular weight is 16 kDa. Use a 12-15% SDS-PAGE gel and optimize transfer conditions for low molecular weight proteins.
- Immunohistochemistry (paraffin) may require antigen retrieval; mitochondrial targets often benefit from heat-induced epitope retrieval and permeabilization.
- ELISA applications can be developed, but titration and specificity checks are recommended.
