Biological Background: MRP4/ABCC4 Function and Localization
- MRP4 (ABCC4) is a member of the ATP-binding cassette (ABC) transporter superfamily, subfamily C, functioning as an ATP-dependent efflux pump for a wide variety of substrates. Related references: PMID:11856762, PMID:12523936, PMID:15364914
- It transports endogenous signaling molecules, including cyclic nucleotides (cAMP, cGMP), bile acids, steroid conjugates, urate, and prostaglandins. Related references: PMID:11856762, PMID:12523936, PMID:12835412
- The protein mediates efflux of glutathione conjugates like leukotriene C4 (LTC4) and leukotriene B4 (LTB4), with GSH required for LTB4 but not LTC4 transport. Related references: PMID:17959747
- MRP4 facilitates the cotransport of bile acids with reduced glutathione (GSH). Related references: PMID:12523936, PMID:12883481, PMID:16282361
- It handles a wide range of drugs and their metabolites, including anticancer, antiviral, and antibiotic agents, contributing to drug resistance. Related references: PMID:11856762, PMID:12105214, PMID:15454390
- MRP4 confers resistance to anticancer agents such as methotrexate. Related references: PMID:11106685
- Subcellular localization includes both basolateral and apical cell membranes, depending on tissue context.
- Tissue specificity: widely expressed, with particularly high levels in prostate, barely detectable in liver, and present at the sinusoidal membrane of hepatocytes.
Experimental Guidance and Technical Tips
- The immunogen covers amino acids 1067–1325, which includes part of the nucleotide-binding domain 2 and the C-terminal region; it may recognize full-length MRP4 in denaturing conditions.
- For Western blot, the predicted molecular weight is ~150 kDa; consider using a 6–8% SDS-PAGE gel for optimal resolution.
- In IHC-P, antigen retrieval with citrate buffer (pH 6.0) or EDTA (pH 9.0) might be necessary; validate with appropriate positive and negative tissue controls.
- For IF/ICC, fixation with 4% PFA and permeabilization with 0.1% Triton X-100 is typically used; test antibody dilutions starting at 1:50–1:200.
- ELISA can be employed for quantitative detection; optimize coating antigen and antibody concentration according to your specific assay format.
