Biological Background: MRP2/ABCC2 Function and Localization
- MRP2/ABCC2 is an ATP-binding cassette (ABC) transporter that uses ATP hydrolysis to actively export endogenous and xenobiotic substrates conjugated with glucuronate, glutathione, or sulfate from cells. Related references: PMID:10220572 PMID:10421658 PMID:11500505
- It localizes to the apical membrane of polarized cells, including hepatocytes, renal epithelial cells, and intestinal epithelial cells, where it mediates the hepatobiliary and renal excretion of conjugated compounds, preventing their reabsorption into the bloodstream.
- The transporter is critical for canalicular bile production and bilirubin detoxification by mediating the excretion of mono- and bis-glucuronidated bilirubin. Related references: PMID:10421658
- MRP2/ABCC2 exhibits low affinity but high transport velocity for steroid glucuronates, including testosterone, dihydrotestosterone, estradiol, estrone, and estriol glucuronates, potentially regulating their metabolic clearance. Related references: PMID:29175180 PMID:30959153 PMID:31704245
- It facilitates the efflux of inflammatory leukotriene C4 (LTC4) and related cysteinyl leukotrienes, and transports sulfated bile salts such as taurolithocholate sulfate, though it is inactive toward androgen sulfates. Related references: PMID:11500505 PMID:16332456 PMID:39993711
- MRP2/ABCC2 contributes to multidrug resistance by transporting various anticancer agents, including anthracyclines, vinca alkaloids, methotrexate, and HIV protease inhibitors. Its expression in tumor cells can reduce intracellular drug accumulation, conferring resistance to cisplatin, doxorubicin, epirubicin, etoposide, and vincristine. Related references: PMID:10220572 PMID:11500505 PMID:12441801
- Expressed primarily in polarized cells of the liver, kidney, and intestine, with highest levels in the liver; also detected in small intestine. Related references: PMID:28408210
Experimental Guidance and Technical Tips
- The immunogen corresponds to amino acids 180–315 within the N-terminal region of human MRP2/ABCC2; this region may confer epitope specificity. Researchers should assess cross-reactivity with related ABCC family members.
- For Western blotting, the predicted molecular weight is ~174 kDa; use a lower percentage polyacrylamide gel (e.g., 6–8%) for optimal resolution and ensure adequate transfer time for high-molecular-weight proteins.
- In immunofluorescence/ICC, the protein localizes to the apical membrane; appropriate permeabilization and fixation conditions should be optimized. Co-staining with apical markers is recommended.
- For ELISA, the antibody can be used as capture or detection reagent; titer and optimal working concentration must be determined empirically using the target antigen.
- The antibody cross-reacts with mouse and rat MRP2/ABCC2 as specified, but sequence alignment of the immunogen region is advised to confirm epitope conservation across species.
