Biological Background: MRP1/ABCC1 Function and Localization
- MRP1/ABCC1, encoded by the ABCC1 gene, is an ATP-binding cassette sub-family C member 1 that functions as a multidrug resistance-associated protein. It is also known as leukotriene C4 transporter and GS-X pump.
- The protein functions as an ATP-dependent efflux pump, exporting a broad spectrum of organic anions, glutathione conjugates, and xenobiotics such as leukotriene C4, estradiol-17-beta-o-glucuronide, and methotrexate, and hydrolyzes ATP with low efficiency. - MRP1 confers resistance to anticancer agents by reducing intracellular drug accumulation through ATP- and GSH-dependent export. Related references: PMID:9281595

- The immunogen is a recombinant fragment covering amino acids 850-960 of human MRP1/ABCC1 (NP_004987.2). This region may present linear epitopes suitable for detection in denaturing conditions.
- For western blot (WB), anticipate a major band at approximately 172 kDa. Use appropriate loading controls and validate in human-derived samples.
