Biological Background: MRM3 Function and Localization
- MRM3 (rRNA methyltransferase 3, mitochondrial), also known as RNMTL1 and RMTL1, is a mitochondrial protein identified through proteomics and is a member of the RNA methyltransferase family.
- It functions as an S-adenosyl-L-methionine-dependent 2'-O-ribose methyltransferase that specifically catalyzes the formation of 2'-O-methylguanosine at position 1370 (Gm1370) in the 16S mitochondrial large subunit ribosomal RNA (mtLSU rRNA), a critical modification located in the peptidyl transferase domain. Related references: PMID:24036117, PMID:25009282, PMID:25074936
- Additionally, MRM3 is required for the formation of 2'-O-methyluridine at position 1369 (Um1369), a modification mediated by the enzyme MRM2. Related references: PMID:35177605
- The protein localizes exclusively to the mitochondrion and possesses a transit peptide that directs it to this organelle.
- MRM3 is associated with the keywords: 3D-structure, Methyltransferase, rRNA processing, S-adenosyl-L-methionine, and Transferase.
- Tissue specificity data indicates that MRM3 is expressed at comparable levels in both normal liver tissue and hepatocarcinoma.
Experimental Guidance and Technical Tips
- The immunogen corresponds to the full-length human MRM3 protein (amino acids 1-420), which is predicted to yield polyclonal antibodies recognizing multiple epitopes along the entire sequence, including the mature and precursor forms.
- For Western blotting, consider that the theoretical molecular weight of the full-length protein is approximately 47 kDa; however, cleavage of the mitochondrial transit peptide may result in a mature protein of slightly lower observed mass. Validate the detected bands in relevant mitochondrial-enriched samples.
- In ELISA applications, the antibody may detect native and recombinant MRM3; appropriate controls (e.g., antigen-coated vs. control wells) should be included to confirm specificity.
