Biological Background: MRE11 Function and Localization
- MRE11 is a core component of the MRN complex, essential for double-strand break repair, DNA recombination, telomere maintenance, and meiosis. Related references: PMID:11741547, PMID:14657032, PMID:22078559
- The MRN complex facilitates homologous recombination repair by mediating DNA end resection. MRE11 first endonucleolytically cleaves the 5' strand to prevent NHEJ, then generates 3' single-stranded DNA via exonuclease activity, enabling long-range resection by EXO1 and DNA2. - MRE11 possesses both single-strand endonuclease and double-strand-specific 3'-5' exonuclease activities. - The immunogen corresponds to amino acids 290-485 of human MRE11, a central region. This polyclonal antibody is expected to detect full-length MRE11 (81 kDa) in denaturing western blot (WB). Optimize the primary antibody dilution (e.g., 1:500-1:2000) for your experimental system.
- For immunoprecipitation (IP), consider using at least 2-5 µg of antibody per 500 µg of cell lysate. Include appropriate positive and negative controls, and validate the target band by mass spectrometry or siRNA knockdown if needed.
- In ELISA applications, titrate the antibody against the immunogen or purified protein to determine the optimal working concentration. Confirm specificity by testing against unrelated antigens.
- Since cross-reactivity is reported for human and rat, validate the antibody in your species and tissue of interest using positive controls known to express MRE11.
