- MRC2, also known as CD280, ENDO180, UPARAP, or CLEC13E, is a type I transmembrane C-type lectin receptor (Gene ID: 9902) involved in endocytosis and extracellular matrix remodeling.
- It functions as a calcium-dependent endocytotic lectin receptor, internalizing glycosylated ligands via clathrin-mediated endocytosis and directing them to endosomal compartments.
- MRC2 may contribute to the regulation of the plasminogen activation system by controlling extracellular levels of PLAUR/PLAU, thereby modulating cell-surface protease activity.
- The receptor plays a key role in collagen turnover by mediating the uptake, remodeling, and degradation of extracellular collagen matrices, often in cooperation with matrix metalloproteinases (MMPs). This function implicates MRC2 in cancer progression and chronic tissue-destructive diseases.
- MRC2 is ubiquitously expressed at low levels in most adult tissues, including brain, placenta, lung, kidney, pancreas, and colon, with higher expression in fetal lung and kidney. It is predominantly found in endothelial cells, fibroblasts, and macrophages.

- The immunogen (amino acids 120-300 of human MRC2) maps to the extracellular region, suggesting the antibody recognizes the receptor's extracellular domain. For Western blot, consider a starting dilution range of 1:500–1:2000 under reducing conditions, and expect a band near 167 kDa. Validate using positive control lysates from cell lines such as HeLa, human fibroblasts, or macrophages.
- For ELISA applications, use the recombinant immunogen as a standard and optimize coating and detection conditions empirically. The antibody's cross-reactivity with mouse MRC2 enables its use in mouse model studies; however, users should independently verify reactivity in their specific mouse strain or tissue.
