Biological Background: Mras (Ras-related protein M-Ras / R-Ras3) Function and Localization
- Ras-related protein M-Ras (also known as R-Ras3) is encoded by the MRAS gene in humans and belongs to the Ras family of small GTPases.
- Acts as a signal transducer in the Ras-MAPK signaling pathway, thereby regulating cell proliferation and survival. Related references: PMID:16630891 PMID:28289718 PMID:35768504
- When GTP-bound, serves as a core component of the SHOC2-MRAS-PP1c (SMP) holophosphatase complex, which specifically dephosphorylates inhibitory serine residues on RAF kinases (RAF1 Ser-259, BRAF Ser-365, ARAF Ser-214) to stimulate their kinase activities. Related references: PMID:16630891 PMID:35768504 PMID:35830882
- Unlike other Ras proteins, MRAS has low intrinsic GTPase activity and may require additional factors for efficient activation. Related references: PMID:39809765
- Recognized by the LZTR1 adapter for ubiquitination by a BCR (BTB-CUL3-RBX1) E3 ubiquitin-protein ligase complex, providing a layer of regulatory control. Related references: PMID:40934300
- Localized primarily at the cell membrane, consistent with its role in relaying extracellular signals to intracellular kinases.
- Expression is highly restricted to the brain and heart, suggesting specialized functions in these tissues.
- Contains multiple regulatory features including alternative splicing, prenylation, and methylation; associated with disease variants.
Experimental Guidance and Technical Tips
- The immunogen corresponds to a central region of human Mras (amino acids 109-208); consider verifying antibody specificity via peptide blocking or siRNA/shRNA-mediated knockdown/knockout controls.
- For Western blot, a band around 24 kDa is expected under reducing conditions; use appropriate molecular weight markers and validate in relevant cell or tissue lysates.
- This antibody is recommended for ELISA; optimal coating buffer and antibody concentrations should be determined empirically for your specific assay format.
- Cross-reactivity with mouse and rat proteins is indicated by the supplier; independent validation in the intended sample system is advised.
