Biological Background: MPI Function and Localization

  • MPI (Mannose-6-phosphate isomerase), also known as phosphomannose isomerase (PMI) or phosphohexomutase, is encoded by the MPI gene in humans. Alternative splicing produces multiple transcript variants.
  • It functions as a key isomerase that catalyzes the reversible interconversion of fructose-6-phosphate and mannose-6-phosphate, a critical step in supplying D-mannose derivatives for eukaryotic glycosylation pathways.
  • The enzyme is a zinc-dependent metalloisomerase and belongs to the mannose-6-phosphate isomerase family.
  • Subcellularly, MPI localizes predominantly to the cytoplasm.
  • It is ubiquitously expressed in all tissues, with relatively higher abundance in heart, brain, and skeletal muscle.
  • Post-translational modifications include acetylation and phosphorylation, which may regulate its activity or stability.
  • Genetic defects in MPI are associated with congenital disorder of glycosylation type Ib (CDG1B), highlighting its essential role in normal glycosylation.

Experimental Guidance and Technical Tips

  • This rabbit polyclonal antibody was generated using a full-length recombinant MPI protein immunogen; it is expected to detect endogenous MPI in human, mouse, and rat samples by WB and ELISA.
  • For Western blotting, a band around 47 kDa corresponding to MPI should be observed; optimizing primary antibody dilution and blocking conditions is recommended for each experimental system.
  • In ELISA applications, the antibody can be used as a capture or detection reagent; pair it with a suitable secondary antibody for colorimetric or chemiluminescent readout.
  • Since the immunogen spans the entire protein, the antibody may recognize multiple isoforms if present; confirm specificity by using controls such as knockout cell lines or recombinant protein.
