- CD31/PECAM1 is an immunoglobulin superfamily cell adhesion molecule essential for leukocyte transendothelial migration (TEM). Tyr-679 is critical for trafficking to the lateral border recycling compartment (LBRC).
- It mediates homophilic endothelial cell-cell adhesion at cell junctions, contributing to vascular barrier integrity.
- Heterophilic binding to CD177 facilitates neutrophil transendothelial migration.
- Homophilic ligation of PECAM1 on viable leukocytes transmits a detachment signal that prevents macrophage-mediated phagocytosis, while on apoptotic cells this signal is disabled, promoting tethering and clearance.
- PECAM1 modulates bradykinin receptor BDKRB2 activation and regulates ERK1/2 signaling in endothelial cells.
