Biological Background: CD31/PECAM1 Function and Localization

  • CD31/PECAM1 is an immunoglobulin superfamily cell adhesion molecule essential for leukocyte transendothelial migration (TEM). Tyr-679 is critical for trafficking to the lateral border recycling compartment (LBRC).
  • It mediates homophilic endothelial cell-cell adhesion at cell junctions, contributing to vascular barrier integrity.
  • Heterophilic binding to CD177 facilitates neutrophil transendothelial migration.
  • Homophilic ligation of PECAM1 on viable leukocytes transmits a detachment signal that prevents macrophage-mediated phagocytosis, while on apoptotic cells this signal is disabled, promoting tethering and clearance.
  • PECAM1 modulates bradykinin receptor BDKRB2 activation and regulates ERK1/2 signaling in endothelial cells.
  • It has been implicated in atherosclerosis susceptibility in mice. Related references: PMID:19048083
  • The protein localizes to cell membrane rafts and cell junctions, and undergoes post-translational modifications including palmitoylation, phosphorylation, and N-glycosylation.
  • Expressed in lung vasculature and platelets.

Experimental Guidance and Technical Tips

  • For flow cytometry, use live, unfixed cells to preserve surface CD31 expression; the immunogen (aa 18-220) spans the extracellular domain, suggesting the antibody binds an extracellular epitope.
  • Titrate the antibody to determine the optimal staining concentration for your cell type of interest (e.g., mouse endothelial cells or platelets).
  • Include appropriate isotype controls and single-stain compensation controls when performing multicolor flow cytometry.
  • Validate cross-reactivity in rat samples if applicable, as the antibody is reported to recognize rat CD31.
