Biological Background: IL4 Function and Localization
- Interleukin-4 (IL4, also known as B-cell stimulatory factor 1/BSF-1) is a secreted cytokine that plays a key role in regulating antibody production, hematopoiesis, inflammation, and the development of effector T-cell responses. Related references: PMID:3083412
- IL4 induces the expression of class II MHC molecules on resting B-cells and enhances both secretion and cell surface expression of IgE and IgG1. Related references: PMID:3498301

Experimental Guidance and Technical Tips
- For intracellular flow cytometry (FC intra), cells must be properly fixed and permeabilized to allow antibody access to the secreted IL4. Use of protein transport inhibitors (e.g., brefeldin A) may be required to accumulate intracellular cytokine.
- The immunogen corresponds to amino acids 21-140 of mouse IL4 (NP_067258.1). Validate antibody specificity using IL4 knockout mouse cells or by pre-incubating with recombinant mouse IL4 protein as a blocking control.
- This antibody is intended for mouse samples; cross-reactivity with other species has not been tested. Consider confirming performance in your experimental system with appropriate positive (e.g., activated Th2 cells) and negative controls.
