- Interleukin-1 receptor type 1 (IL-1R1, CD121a) is a transmembrane receptor encoded by the Il1r1 gene, and serves as the primary receptor for interleukin-1 (IL-1) cytokines IL1A, IL1B, and the IL-1 receptor antagonist IL1RN.
- Upon ligation, IL-1R1 associates with the coreceptor IL1RAP to form a high-affinity signaling complex, triggering downstream NF-kappa-B and MAPK pathways critical for inflammatory and immune responses.
- The receptor's intracellular TIR domain recruits adaptors like MYD88 and IRAKs, leading to kinase cascades; IL-1R1 also participates in IL1B-mediated costimulation of IFNG production in Th1 cells.
