Biological Background: IL-33R/ST2 Function and Localization

  • IL-33R/ST2 (Interleukin-1 receptor-like 1) is the receptor for interleukin-33 (IL-33), playing essential roles in innate and adaptive immunity, tissue homeostasis, and responses to environmental stresses. Related references: PMID:17675517, PMID:18450470, PMID:22660580
  • Upon IL-33 binding, ST2 recruits the coreceptor IL1RAP and downstream adaptors MYD88, IRAK1/4, and TRAF6, leading to phosphorylation of MAP kinases ERK1/2 and p38 MAPK.
  • ST2 signaling induces UCP2-dependent mitochondrial rewiring upon tissue injury, attenuating reactive oxygen species generation and preserving Krebs cycle integrity to promote itaconate production and GATA3-dependent differentiation of anti-inflammatory macrophages.   • The receptor exists as multiple isoforms via alternative splicing: a long membrane-bound form (ST2L) and a soluble secreted form (sST2), which acts as a decoy inhibitor of IL-33 signaling.
  • ST2 is predominantly expressed in hematopoietic tissues, including macrophage, erythroid, epithelial, and fibroblast cell lines; isoform A is found in brain astrocytes and microglia, isoform B in brain endothelial cells.
