Biological Background: CD98 (SLC3A2) Function and Localization
- CD98 heavy chain, also known as SLC3A2 or 4F2hc, is a type II transmembrane glycoprotein that acts as a chaperone for amino acid transporter light chains of the SLC7 family, forming functional heterodimers essential for amino acid transport (PubMed:10574970, PubMed:10734121, PubMed:11011012). Related references: PMID:10574970 PMID:10734121 PMID:11011012
- These heterodimers, such as SLC3A2/SLC7A5 (LAT1), mediate the cellular uptake of large neutral amino acids including leucine, which in turn activates mTORC1 signaling upon lysosomal recruitment by LAPTM4B.
- SLC3A2 modulates integrin signaling and is crucial for integrin-dependent cell spreading, migration, and tumor progression (By similarity).
- The protein is broadly expressed in mouse tissues, with highest levels in kidney, small intestine, spleen, thymus, and liver; it is also detected on brain capillary endothelia and embryonic epithelia (PubMed:9915839). Related references: PMID:9915839
- Subcellularly, SLC3A2 localizes to the plasma membrane (both apical and basolateral surfaces), lysosomes, melanosomes, and cell junctions, reflecting its diverse roles in transport and adhesion.
- It undergoes various post-translational modifications including N-glycosylation, disulfide bond formation, phosphorylation, and ubiquitination, and is subject to alternative splicing giving rise to multiple isoforms.
Experimental Guidance and Technical Tips
- Since this antibody is directly conjugated to ABflo 647, no secondary antibody is needed for flow cytometry, reducing background and experimental steps.
- Titrate the antibody to determine the optimal staining concentration for your mouse cell samples, and include appropriate isotype controls and viability dyes.
- The immunogen spans the extracellular domain (amino acids 105-526), so the antibody should recognize the native surface form of CD98 on live cells.
- Validate the antibody in the relevant mouse tissue or cell line, considering that CD98 expression may vary; positive controls could include activated lymphocytes or tumor cell lines.
